pTlpA ccdA-B
pTIpA, regulated by TlpA, is upstream the ccdA antitoxin. When temperature goes below 37 °C, this leads to a decreased expression of the antitoxin and the toxin ccdB leads to the microorganism’s death.
This construct consists of the toxin ccdB with the placUV5 promoter (BBa_K4244018) which is reversed, together with pTlpA and the antitoxin ccdA (BBa_K4244016) and the TlpA (BBa_K4244017). This construct has been tested in E.coli Nissle 1917 with a temperature sensitive survival assay. Unfortunately, the experiment did not show the expected results (decrease of colony number with decrease of temperature).
